El desarrollo tumoral y la metástasis son dos procesos que están íntimamente ligados al
desarrollo de la vasculatura. Al igual que cualquier órgano, los tumores necesitan crear
un sistema vascular que colme sus necesidades metabólicas pero para poder desplegar
esta nueva red vascular requieren de la modificación de la matriz extracelular (ECM,
extracelular matrix). La proteólisis de la ECM se lleva a cabo por un grupo de proteínas
con capacidad proteolítica llamadas metaloproteasas de la matriz. Esta familia de
proteasas se divide a su vez en tres familias denominadas MMPs, ADAMSs y
ADAMTSs. Dentro de la familia ADAMTSs, se ha descrito que el primero de sus
miembros es capaz de regular el desarrollo tumoral a través de un mecanismos que
influye en la formación de nuevos vasos sanguíneos. A su vez, se ha descrito que
ADAMTS1 es necesario para uno de los procesos alternativos de vascularización
tumoral, el mimetismo vasculogénico.
En nuestro trabajo nos centramos en estudiar el papel de ADAMTS1 como modulador
de la angiogénesis y, a su vez, como regulador del desarrollo vascular. Para ello
desarrollamos tumores singénicos mediante la inyección de células de melanoma
murino B16F1 en una cepa de ratón C57Bl/6 ADAMTS1 KO. En este contexto
observamos que ADAMTS1 actuaba como molécula antiangiogénica aunque promovía
el desarrollo tumoral. Esto se debe a que ADAMTS1 disminuye el número de vasos en
los tumores desarrollados en ratones ADAMTS1 WT pero promueve la maduración de
éstos aumentando su tamaño y su funcionalidad. Posteriormente, tras la inhibición de
ADAMTS1 en las células tumorales pudimos comprobar que el efecto de que el
ADAMTS1 derivado de los tumores tenía sobre el desarrollo de la vasculatura tumoral
era limitado o nulo.
Al mismo caracterizamos la capacidad de formación de redes similares a endoteliales
por parte de las células B16F1 y encontramos que ADAMTS1 participa en la formación
de estás redes, aunque de manera limitada.
