Para este tipo de cáncer, la enzima metaloproteasa 9 (MMP-9) ha sido investigada como biomarcador en numerosos estudios, debido a que presenta una elevada expresión en el tejido mamario tumoral. Este hecho se debe a que MMP-9 está involucrada en diferentes etapas de la carcinogénesis como es la proliferación tumoral, la angiogénesis y la invasión y metástasis. Por otra parte, los diferentes tipos de tratamiento estándar para las pacientes de cáncer de mama, entre los que se encuentran la cirugía, la radioterapia, la quimioterapia, la terapia con hormonas y la terapia dirigida, están evolucionando actualmente gracias a la nanomedicina. Por tanto, este estudio se centra en analizar la potencialidad del uso de MMP-9 en las líneas celulares MCF-7, MDA-MB-231 y 4T1 del cáncer de mama mediante la nanotecnología. Para ello, se ha optado por tres enfoques diferentes. El primero utiliza la MMP-9 para dirigir a las nanopartículas hacia el tumor y, así, poder detectar y evaluar el cáncer de mama mediante técnicas de imagen. Aunque el segundo planteamiento vuelve a utilizar a esta enzima como guía, también la emplea para administrar el fármaco incorporado a la nanopartícula en el interior del tumor. Por último, se analiza la utilización de nanopartículas para inhibir la actividad de la enzima MMP-9 en las diferentes etapas de la carcinogénesis en las que participa. Para la inhibición de esta enzima no solo se ha considerado las nanopartículas que tienen a MMP-9 como diana, sino también aquellas que bloquean algún factor de la ruta metabólica en la que esté involucrada. Se ha comprobado que tanto los nanosensores como los nanotransportadores dirigidos por MMP-9 son bastante eficaces, aunque no tienen una elevada especificidad entre las gelatinasas. Por otro lado, los tratamientos con nanopartículas que regulan a MMP-9 como diana han demostrado tener una buena eficacia, considerando la citotoxicidad que produce en el organismo. La revisión realizada permite concluir que esta nueva técnica presenta resultados prometedores en la actualidad y plantea la necesidad de investigar con mayor profundidad para poder aplicarla a la práctica clínica. Palabras claves: cáncer de mama, MMP-9, nanomedicina, nanopartículas. ABSTRACT Nowadays, breast cancer is the second most widespread cancer worldwide and the most frequent in women. For this type of cancer, the metalloprotease enzyme 9 (MMP-9) has been investigated as a biomarker in numerous studies, due to the facts its presents a high expression in the mammary tumor tissue. This is due to the fact that MMP-9 is involved in different stages of carcinogenesis such as tumor proliferation, angiogenesis and invasion and metastasis. Furthermore, the types of standard treatment for cancer patients, as radiotherapy, chemotherapy, hormone therapy and targeted therapy, are currently evolving thanks to nanomedicine. Therefore, this research focuses on analyzing the potential use of MMP-9 in breast cancer cell lines MCF-7, MDA-MB-231 and 4T1 through nanotechnology. In order to do this, three different approaches have been chosen. The first uses MMP-9 to direct the nanoparticles to the tumor, and thus, to detect and evaluate breast cancer using imaging techniques. Although the second approach uses this enzyme as a guide as well, it also employs it to administer the drug incorporated into the nanoparticle into the tumor. Finally, the use of nanoparticles is analyzed to inhibit the activity of the MMP-9 enzyme in the different stages of carcinogenesis in which it participates. For the inhibition of this enzyme not only have they been considered both the nanoparticles that have MMP-9 as a target, but also those which block any factor of the metabolic pathway in which it may be involved. It has been confirmed that both nanosensors and nanotransporters driven by MMP-9 are quite effective, in spite of not having a high specificity among gelatinases. On the other hand, the treatments with nanoparticles that regulate MMP-9 as a target have been proven to have a good efficacy, considering the cytotoxicity it produces in the organism. The review carried out allows us to conclude that this new technique presents promising results nowadays, and raises the need to investigate in greater depth to be able to apply it to clinical practice.
