dc.description.abstractEl gen que codifica el antígeno de superficie más abundante de Leishmania infantum, la Gp63, ha sido clonado y secuenciado. A pesar de la homología general que se encuentra al compararlo con genes de otras especies de Leishmania, en particular con el gen expresado constitutivamente por Leishmania ehagasi, los extremos correspondientes al carboxilo terminal de la proteína son claramente divergentes (62% homología). Para estudiar la predominancia de los anticuerpos anti-Gp63 en sueros de perros con leishmaniasis visceral, se clonó y expresó Gp63 recombinante en Eseheriehia eolio El 100% de dichos sueros reconoció la proteína recombinante sugiriendo que podría funcionar como un potente inmunógeno de células B durante la leishmaniasis visceral canina. Sin embargo, se observó heterogeneidad en el nivel de respuesta de los sueros. Mediante el uso de 6 fragmentos solapantes de la Gp63 y de una colección de péptidos sintéticos se analizaron sus determinantes antigénicos. Los datos muestran que debe existir un cierto grado de restricción inmunológica en el reconocimiento de la proteína ya que se observa una reactividad preferente con la región más divergente de la misma. El mapeo de epitopos de esta región mostró la existencia de dos péptidos inmunodominantes y la respuesta inmunológica producida contra ellos es preferencialmente del tipo IgG2a, isotipo asociado a la respuesta de tipo Thl.es_ES
dc.description.abstractThe Gp63 gene encoding the major surface antigen of Leishmania infantum has been cloned and sequenced. In spite of the overall sequence homology with the gp63 genes from other Leishmania species, particularly with the constitutively expressed Leishmania ehagasi Gp63 gene, the 3'ends of these genes are clearly divergent (62% homology). To study tbe prevalence of anti-gp63 antibodies in the sera from dogs with visceralleishmaniasis, a recombinant L. infantum gp63 protein was expressed in Eseheriehia eolio 100% of the sera from tbese dogs recognized the recombinant gp63 protein suggesting that it must function as a potent B cell immunogen during natural canine visceral leishmaniasis. However, heterogeneity in the level of response was observed. Fine mapping of the antigenic determinants was performed by means of 6 overlapping subfragments of the gp63 protein and by the use of a library of synthetic peptides. The data showed that there is some degree of immunological restriction in the recognition of the protein since preferential reactivity was observed against the most divergent region, Epitope mapping of this region showed two immunodominant peptides the response to wruch is preferentially ofthe Ig02a type, isotype associated to Thl response.es_ES
