Se ha desarrollado un método fluorimétrico rápido y sensible para la estimación del clorhidrato de doxorubicina
(DH) en sangre entera y tejidos de ratas. También se ha desarrollado un método relativamente más simple y sensible
para la estimación en plasma. Los parámetros analíticos se establecieron y compararon en ambos métodos. Los datos
de intensidad de fluorescencia relativa (IFR) de los métodos de estimación de DH en plasma y en sangre entera
indican la elevada sensibilidad del método anterior. La comparación de los valores de IFR de la sangre entera y de
los tejidos indican una baja extracción de DH de la sangre entera. Los valores bajos de IFR se observaron en tejidos
que contienen elementos formadores de sangre, tales como el bazo y el hígado, mientras que en pulmones, riñones
y corazón los valores de IFR detectados fueron elevados. La incubación de DH con diversas concentraciones de ADN
in vitro mostró una reducción de la IFR del DH en comparación con la IFR inicial, lo que indica una fuerte
interacción dependiente de la concentración. Se observaron coeficientes de correlación elevados en pulmones (0,9996),
sangre entera (0,9985) y corazón (0,9991). La prueba de interceptación realizada para el método de estimación de
DH en plasma y el método de estimación en sangre entera y tejidos indica que no se producen interferencias de
blancos en las estimaciones. La prueba ‘t’ realizada para las concentraciones estimadas en los estudios de recuperación
no indican ninguna diferencia significativa entre las concentraciones añadidas y estimadas que demuestran la exactitud, y el bajo coeficiente de variación (%) y los límites de confianza indican la precisión de los métodos. A rapid and sensitive fluorimetric method of estimation of doxorubicin hydrochloride (DH) in whole blood and tissues
of rat was developed. A relatively simpler and sensitive method of estimation in plasma was also developed. The
analytical parameters were established for both the methods and were compared. The data of relative fluorescence
intensities (RFI) of the methods of estimation of DH in plasma and whole blood indicate high sensitivity of the former
method. Comparison of RFI values of whole blood and tissues indicate low extraction of DH from whole blood. Low
RFI values were observed in case of tissues such as spleen and liver containing blood forming elements, where as, high
RFI values were found in case of lung, kidney and heart. Incubation of DH with various concentrations of DNA in vitro,
showed a decrease in RFI of DH with time compared to the initial RFI, indicating a strong interaction which is
concentration dependant. High correlation coefficients were observed with lung (0.9996), whole blood (0.9985) and
heart (0.9991). The test of intercept performed for the method of estimation of DH in plasma, and the method of
estimation in whole blood and tissues indicate no interference of blank in the estimations. The ‘t’ test performed for the
estimated concentrations in recovery studies indicate no significant difference between the added and estimated concentrations
proving the accuracy, and low coefficient of variation (%) and confidence limits indicate the precision of the methods.
